Abstract: ถั่วเน่า เป็นอาหารหมักที่ชาวบ้านทางภาคเหนือตอนบนของไทยนิยมรับประทานในครัวเรือนและทำเป็นผลิตภัณฑ์จำหน่ายตามท้องตลาด โดยปกติแล้วชาวบ้านทั่วไปนิยมใช้วิธีการหมักถั่วเน่าแบบภูมิปัญญาท้องถิ่น ถั่วเน่าที่ได้จึงมีรสชาติและลักษณะไม่สม่ำเสมอ ดังนั้นการคัดแยกเชื้อจากถั่วเน่าให้เป็นเชื้อบริสุทธิ์ เพื่อเป็นเชื้อสำเร็จรูปและหาสูตรต่างๆ ที่เหมาะสมสำหรับการหมักถั่วเน่าจึงช่วยคงคุณภาพถั่วเน่าให้มีคุณภาพที่ดีได้ โดยทำการแยกเชื้อบริสุทธิ์จากถั่วเน่าที่ผลิตในพื้นที่ตำบลขี้เหล็ก อำเภอแม่ริม จังหวัดเชียงใหม่ ที่ผ่านกระบวนการหมักเป็นเวลา 2 และ 3 วัน (บดละเอียดและเมล็ด) และศึกษาค่าความเป็นกรด-ด่าง (pH) จำนวนเชื้อเริ่มต้น การผลิตเอนไซม์โปรติเอส และการประเมินความชอบ (sensory test) โดยหมักถั่วเน่าแผ่นจำนวน 8 สูตร ได้แก่ Control, A, AB, AC, AD, AE, C, CE ระยะเวลา 24 48 และ 72 ชั่วโมง จากการทดลองพบว่า สามารถแยกเชื้อจากถั่วเน่าได้ 5 ไอโซเลต สภาวะที่เหมาะสมต่อการหมักถั่วเน่าอยู่ในชั่วโมงที่ 48 โดยค่า pH แปรผันตามระยะเวลาที่ทำการหมัก จากการคุณสมบัติของเอนไซม์ โปรติเอสโดยวิธี azocasein พบว่ากิจกรรมเอนไซม์โปรติเอสอยู่ระหว่าง 0.25-3.29 unit/ml การประเมินความชอบโดยรวมของถั่วเน่าแผ่น พบว่าชาวบ้านชอบสูตร AB มากที่สุด ต่อมาคือสูตร control, A, AE, AC, CE, C และ AD ตามลำดับ โดยมีระดับคะแนนอยู่ที่ 22.70±1.60, 22.57±2.20, 22.50±2.08, 21.60±2.82, 21.20±2.46, 21.20±3.20, 21.00±3.25 และ 18.23±4.71 ตามลำดับ โดยมีความแตกต่างกันอย่างมีนัยสำคัญทางสถิติ ที่ความเชื่อมั่น 95 เปอร์เซ็นต์
Description: Thua nao is the Thai fermented soybean product. The people of northern Thailand on the popular and made household products sold by the market. Typically, people used more common fermented Thua nao as local knowledge that is not consistent with taste and style. Therefore, the bacteria isolated from infected Thua nao into pure infection and is ready to find formulas. Suitable for fermentation of Thua nao is to preserve the quality of good quality. Purification of Thua nao produced in Khi Lek, Mae Rim, Chiang Mai. The fermentation process for 2 and 3 days (pulp and seeds) and pH, inoculums, protease and sensory test with fermented Thua nao of 8 formula Control, A, AB, AC, AD, AE, C and CE at 24, 48 and 72 hours showed that Isolates from the five Thua nao isolate the optimal conditions for fermentation, Thua nao in 48 minutes by varying the pH of fermentation Period. From the properties of the enzyme activity by azocasein showed that protease are between 0.25-3.29 unit / ml to evaluate the overall liking of Thua nao sheet Found that people prefer the formula AB biggest impact was the recipe control, A, AE, AC, CE, C and AD, respectively, with scores were 22.70±1.60, 22.57±2.20, 22.50±2.08, 21.60±2.82, 21.20±2.46, 21.20±3.20, 21.00±3.25 and 18.23±4.71 respectively, with different statistical significance 95 percent confidence.
